Hi,
In Vista, Itunes stores its data by default in c:\users\<usename>\music.
You can change this folder in the configuration menu under edit - preferences - advanced and get it to point to some folder on the external drive. If you do this after first install you are sorted but if you want to move existing data you are in trouble.
In theory, if you move the itunes contents from c:\users\<usename>\music to your external drive beforehand, it should work but you will get issues with duplicated songs in itunes afterwards that have to be manually removed and I am not sure how to avoid this. Frankly, I don't know why they have to make this so difficult and unintuitive. You are probably better off asking the question in one of the apple forums.
